2 材料与方法Materials and Methods
2.1 试剂Chemicals
所有的溶剂与试剂均为色谱级,磺胺嘧啶,磺胺甲恶唑,磺胺二甲基嘧啶,氧氟沙星,诺氟沙星,环丙沙星,氯霉素,甲氧苄氨嘧啶标准品购于xxx
All chemicals and solvent were HPLC. Standard: sulfadiazine, sulfamethoxazole, sulfamethazine, ofloxacin, norfloxacin, ciprofloxacin, chloramphenicol, trimethoprim and were purchased from xxx.
2.2 样品处理Sample Preparation
1) 将样品摇匀,吸取500 mL左右的样品,用稀盐酸调pH至4.2;
2) 通过HLB固相萃取柱萃取,流速控制在 xxx mL/min以下;
3) 样品过柱后,对萃取柱继续抽真空干燥10 min;
4) 用6 mL含5%甲醇的二次水淋洗萃取柱,再用5 mL甲醇洗脱;
5) 将样品用氮气浓缩至0. 1mL以下,加内标;
6) 用水(含0.1% xxx)定容至0.5 mL,并用0.45 µm滤膜过滤,进样5 µL。
2.3 HPLC分析HPLC Analysis
本实验所用的仪器为Agilent 1100型高效液相色谱仪,配备二极管阵列检测器。
色谱柱为xxx C18 (150 mm*3.0 mm, 3.5 μm),流动相A为水(含0.1% xxx),B为xxx,流速为0.25 mL/min,柱温为25℃。
