2 材料与方法Materials and Methods
2.1 试剂Chemicals
所有的溶剂与试剂均为色谱级，α-亚麻酸，亚油酸，棕榈酸，油酸，硬脂酸标准品购于xxx。
All chemicals and solvent were HPLC. Standard: α-Linolenic acid, linoleic acid, palmitic acid , oleic acid, stearic acid and were purchased from xxx.

2.2 样品处理Sample Preparation
1) 将样品摇匀，称取5.0g左右的样品；
2) 5.0g样品中加入75 mL xxx，冷浸30 min，超声振荡30 min，过滤；
3) 重复上述提取过程3次，合并滤液，减压蒸干，得到样品油脂；
4) 称取样品油脂0.2g，加入5% NaOH-xxx溶液4 mL，60℃水浴回流30 min；
5) 冷却，加入14% xxx-xxx甲醇溶液5 mL，回流2 min；
6) 加入4 mL xxx回流5 min，冷却，加入5 mL饱和NaCl溶液，振荡分层；
7) 取上清液1 mL，挥干溶剂，残留物用甲醇定容至1 mL，并用0.22 µm滤膜过滤，进样10 µL。

2.3 HPLC分析HPLC Analysis
本实验所用的仪器为xxx型高效液相色谱仪，配备紫外检测器。
色谱柱为xxx C18 (150 mm*4.6 mm, 5 μm)，流动相为甲醇-xxx=85 : 15，流速为1.0 mL/min，柱温为30℃。
