2 材料与方法Materials and Methods
2.1 试剂Chemicals
所有的溶剂与试剂均为色谱级,玉米素,吲哚乙酸,脱落酸,吲哚丁酸,α-萘乙酸,调吡脲,烯效唑标准品购于xxx
All chemicals and solvent were HPLC. Standard: zeatin, indole-3-acetic acid, abscisic acid, 3-indolebutyric acid, 1-naphthalene acetic acid, forchlorfenuron, uniconazole and were purchased from xxx.
2.2 样品处理Sample Preparation
1) 将样品摇匀,称取5.0g左右的样品;
2) 5.0g样品加入10 mL 80% xxx,超声提取15 min;
3) 抽滤,用5 mL 80% xxx洗涤残渣2次,抽滤;
4) 合并滤液,40℃旋转蒸发仪浓缩;
5) 将浓缩后的水相pH调至7.0,用xxx萃取3次,每次5 mL;
6) 挥干乙酸乙酯,用5%甲醇定容,过SPE-PAK C18固相萃取小柱,4 mL 40%甲醇洗脱;
7) 收集上清液和洗脱液,并用0.45 µm滤膜过滤,进样20 µL。
2.3 HPLC分析HPLC Analysis
本实验所用的仪器为Agilent 1100型高效液相色谱仪,配备紫外检测器。
色谱柱为C18 (250 mm*4.6 mm, 5 μm),流动相A为0.2% xxx,流动相B为xxx,流速为1.0 mL/min,柱温为35℃。
