2 材料与方法Materials and Methods
2.1 试剂Chemicals
所有的溶剂与试剂均为色谱级,桑色素,槲皮素,山奈酚,异鼠李素标准品购于xxx
All chemicals and solvent were HPLC. Standard: morin hydtate, quercetin, kaempferol, isorhamnetin and were purchased from xxx.
2.2 样品处理Sample Preparation
1) 将样品摇匀,称取30.0g左右的样品;
2) 30.0g样品加入100 mL xxx,50℃振荡浸提2 h,补足乙醇,摇匀,10000 r/min离心40s;
3) 吸取5 mL浸提液,加入1 mL xxx(含0.2g xxx/mL),通氮气,封口;
4) 80℃水解2h,加入无水乙醇定容至10 mL,置于-20℃冰箱中冷冻1h;
5) 超声振荡5 min,10000 r/min离心40s,上清液用0.45 µm滤膜过滤,进样10 µL。
2.3 HPLC分析HPLC Analysis
本实验所用的仪器为Waters高效液相色谱仪,配备荧光检测器。
色谱柱为xxx C18 (150 mm*4.6 mm, 3 μm),流动相A为0.1 mol/L xxx,B为甲醇,流速为0.3 mL/min,柱温为40℃。
